When a small blood vessel is injured, a series of reactions is initiated leading to the formation of a blood clot. This seals the vessel preventing further blood loss. Many of the clotting factors involved are enzymes which are activated during this cascade of events. There are three parts of the coagulation cascade; the intrinsic pathway, the extrinsic pathway and the common pathway. The end product of both the intrinsic, and the faster extrinsic pathway, is activated Factor X. This factor initiates a further cascade through the common pathway resulting in the formation of a stable fibrin clot at the site of injury.


Key factors in the clotting mechanism can be measured by Binding Site RID kits. These include Fibrinogen and Factor XIII, as well as protease inhibitors and anticoagulants that localise and limit the clotting process, for example anti-thrombin III (ATIII) and Protein C

Serum protein electrophoresis (SPE or SPEP) can be used for the initial screening of serum (or concentrated urine) for monoclonal immunoglobulins or M-protein and light chains (Bence Jones protein), aiding in the diagnosis of monoclonal gammopathies.

However, optimal detection of paraproteins is achieved by screening serum samples both with SPE or CZE (Capillary Zone Electrophoresis) and Freelite® serum free light chain assays.

Samples exhibiting either abnormal bands by SPE or abnormal levels of free kappa, free lambda or κ/λ ratio by Freelite® should be further investigated by immunofixation and quantitation of other specific serum proteins.

Each SPE kit contains reagents sufficient for 100 tests. Each of the 10 gels can be used to run 10 samples.

IgM Lambda and IgG Kappa monoclonal immunoglobulins on a MINIFIX IFE gel

Immunofixation electrophoresis (IFE) is a two-stage process combining agarose gel electrophoresis with immuno-precipitation. It has replaced IEP (immunoelectrophoresis) as the method of choice for identification and characterisation of monoclonal immunoglobulins in serum, urine and cerebrospinal fluid. IFE allows ease of interpretation, fast results (less than 2 hours) and has good sensitivity.
