Radial Immunodiffusion (RID) Kits
Thousands of laboratories world-wide rely upon a method of protein quantitation that has remained a "Gold Standard" for over 25 years. Radial immunodiffusion (RID), which is based on the complexing of antigen and antibody to produce a visible precipitin ring, is still a reference method to which new methods are compared.
RID remains a reliable method and is particularly useful for difficult samples e.g. that are turbid and for which other methods are inappropriate.
The BINDARID™ range of kits, based on conventional RID technology, is suitable for measuring protein concentrations down to approximately 5mg/mL.
NANORID™ kits use a novel patented technology to enable accurate quantitation of very low concentrations of protein in serum, urine, cerebrospinal fluid and other biological materials. Sensitivities as low as 0.15 mg/mL can be achieved.
Binding Site has a wide range of radial immunodiffusion kits:
- Complement proteins including some functional assays
- Human immunoglobulins including IgG and IgA subclasses
- Coagulation
- Other human serum proteins
- And a Digital RID Plate Reader

Human Complement Proteins
The complement system is a complex series of proteins that act as a cascade. They react in the sequence C1q, C1r, C1s, C4, C2, C3, C5, C6, C7, C8, C9; many requiring proteolytic cleavage to become active. The cascade is activated either via the Classical Pathway, the Alternative Pathway or the Lectin Pathway.
Complement is involved in initiating an inflammatory response and destroying certain bacteria and viruses. Complement proteins are assayed to measure complement consumption and to diagnose inherited deficiencies of cascade components.
Assays for Investigation of Complement Function
Total Haemolytic Complement (THC) and Alternative Pathway Haemolytic Complement (APHC) kits are effective screening assays to aid in the identification of complement deficiency.
The Total Haemolytic Complement kit is primarily designed to detect deficiencies of the Classical Complement Pathway and the terminal sequence (C3-C9) components, whereas the Alternative Pathway Haemolytic Complement kit is designed to measure the activity of the Alternative Complement Pathway.

Secretory IgA
There are two populations of IgA, a monomeric form in serum, and a dimeric form found in secretions such as saliva, tears, bronchial fluids etc. The dimeric form is covalently associated with a J-chain and a secretory piece. This secretory IgA (SIgA) represents the first line of defence against micro-organisms on the mucosal surface. SIgA deficiency results in a lack of immunity to mucosal viruses. This RID kit has been developed to specifically recognise only the SIgA population and not it's component forms (monomeric IgA, J-chain etc).

Human Coagulation Proteins
When a small blood vessel is injured, a series of reactions is initiated leading to the formation of a blood clot. This seals the vessel preventing further blood loss. Many of the clotting factors involved are enzymes which are activated during this cascade of events. There are three parts of the coagulation cascade; the intrinsic pathway, the extrinsic pathway and the common pathway. The end product of both the intrinsic, and the faster extrinsic pathway, is activated Factor X. This factor initiates a further cascade through the common pathway resulting in the formation of a stable fibrin clot at the site of injury.
Key factors in the clotting mechanism can be measured by Binding Site RID kits. These include Fibrinogen and Factor XIII, as well as protease inhibitors and anticoagulants that localise and limit the clotting process - the most significant of these are anti-thrombin III (ATIII), Protein S and Protein C.
